چکیده

Lombok island is a transitional region beetwen Asian and Australian faunal biodiversity. There has been no previous report on the genotype of Helicobacter pylori in the population. The cagA gene is one of the important virulence factors of H. pylori. The diversity of cagA 3′ regions reflect the phylogenetic relationships among the different H. pylori isolates and their association with clinical outcomes and significant geographical differences among isolates have been reported. The purpose of this study was to analyse the variability of 3’region of cagA H. pylori Lombok isolates and determine the genotype based on EPIYA(Glu-Pro-Ile-Tyr-Ala) motif of CagA and its relationship with pathological of the stomach. Total of 36 isolates of H. pylori Lombok origins were studied. Gastric antral biopsies collected from 36 Lomboknese patients (chronic gastritis = 13, gastric/duodenal ulcer =12, and gastric cancer =10) were used to amplify the 3’ regions of cagA containing EPIYA motifs by PCR then followed by DNA sequencing. We have found that 34/36 of the H. pylori Lombok isolates were clustered alone while 1/36 was clustered together with other East Asian strains. The findings of the study suggested that Lombok H. pylori strains share a common ancestry with East Asians but with their own unique sequences. No association of specific sequences with the outcome of disease was revealed through additional phylogenetic analysis based on amino acid sequences.

Phylogenetic analysis of Helicobacter pylori cagA gene of Turkish isolates and the association with gastric pathology

BACKGROUND The cagA gene is one of the important virulence factors of Helicobacter pylori. The diversity of cagA 5' conserved region is thought to reflect the phylogenetic relationships between different H. pylori isolates and their association with peptic ulceration. Significant geographical differences among isolates have been reported. The aim of this study is to compare Turkish H. pylori is...

Presence of cagA Gene in Patients with Gastric Cancer and Gastritis with Helicobacter pylori Infection

Background and purpose: Helicobacter pylori is a gram-negative, micro-aerophilic, and spiral-shaped bacillus. Infection with this bacterium can lead to gastritis, ulcers, and even gastric cancer. This study was conducted to evaluate the presence of the cagA gene among patients with gastritis and gastric cancer. Materials and methods: This study was performed in patients with gastritis (n=40) a...

Analysis of KRAS gene mutation associated with Helicobacter pylori infection in patients with gastric cancer

Objective(s): KRAS proto-oncogene mutation can be considered a diagnostic factor for treating various malignancies. Helicobacter pylori infection, a risk factor for stomach cancer, may cause DNA damage and genetic changes. The aim of the current study was to assess the association of gastric cancer and KRAS mutation, demographic factors, and H. pylori infection.<...

EPIYA motif patterns among Cuban Helicobacter pylori CagA positive strains.

INTRODUCTION It is known that polymorphisms in C-terminal region of CagA influence gastric disease development on Helicobacter pylori infection. Additionally, the geographic distribution of these polymorphisms has been associated with the appearance of more severe gastroduodenal pathologies. Objective. To determine the CagA phosphorylation motifs pattern (EPIYA pattern) in Cuban H. pylori isola...

Helicobacter pylori cagA Status, vacA Subtypes and Histopathologic Findings in Iranian Patients with chronic gastritis

Background and Objective: Helicobacter pylori has several strains with different degrees of virulence. The aim of this study was to detect two major important virulence factors, cagA/vacA genotypes, and to determine correlations among different cagA/vacA genotypes and histological features of chronic gastritis in Iranian patients. Methods: In this cross- sectional study, Gastric bi...
